P. 6128, Succ. A, Montreal, Quebec, H3C 3J7 Physiological Pharmacokinetic (PBPK) models can be used to predict doses of toxic metabolites reaching target tissues. The results of a detailed study of the relative uncertainty and sensitivity associated with individual model parameters in PBPK models for three of volatile organic chemicals: tetrachloroethylene, trichloroethylene and chloroform are presented. Monte Carlo simulations showed that the rate of cardiac output and fractional blood flow to richly perfused tissues and liver were the largest contributors to uncertainty in predicted tissue doses. Predictions were also sensitive to the maximum rate of metabolism Vmax and to the Michaelis-Menten constant Km used to describe liver metabolism, with the degree of sensitivity to Km more pronounced at higher doses. Uncertainty and sensitivity also depend, to a certain extent, on the way in which tissue dose is measured. Here, we considered average daily values of the concentration of the parent compound in the liver, as well as the rate of metabolism and the amount of metabolic product formed.
